Way to unlock the quest: Talk to Rosco.

This quest becomes available right after you have completed the "A Stately Chest" side quest, i.e. after you have located and examined the first chest mentioned by Rosco. Talk to the man again (point 13 on the map) and he will apologize for the mistake. He will then point you to the proper chest. This time, you need to go alone and go East (point 13b on the map). Just like earlier, after you reach the location with the X symbol on the ground, use the shovel and open the chest that you dig up. Also this time, you will not make much profit, because there is only a Hand Mirror inside. Return to Rosco, listen to the conversation and wait for another side quest to become active.
