How to unlock the quest: Through conversation with Burke's Manifestation.

Quest giver: Burke's Manifestation (point 1 on the map).

Get rid of all the Termites.

After reaching a small beach (point 1 on the map), you should be able to notice a friendly Burke (Burke's Manifestation), fighting with a group of Termite Workers. Help Burke out to get rid of the monsters, which shouldn't pose a problem. Remember to use the fact that the Termites will focus their attention on two other characters besides the Nameless Hero (Burke and the actual party member). For each killed Termite you will be given 50 Glory Points, and an additional reward for killing them all - 100 Glory Points.

Speak with Burke and promise to help him.

After the fight end, get closer to Burke (Burke's Manifestation) and speak with him. You will learn from him, that his current form is a result of a failed experiment. You will immediately be asked to join the expedition to the Demon Hunter's Citadel, aimed at restoring Burke's original form.

Note - if you agree to his terms, Burke will temporarily join your party. Don't count on using him in your future fights, however, as he will leave the party immediately should you stray off your path leading to the Demon Hunter's Citadel. You can also order him to wait for the Nameless Hero if you don't want to complete this mission right now.

During your trip to the Citadel, your party may fall onto some spiders.

Depart with Burke to the North, taking a narrow pathway leading towards the areas of the Demon Hunter's Citadel (point 1a on the map). You need to watch out, as your party will surely stumble upon a group of Rock Spiders along the way. The spiders can easily take a huge chunk of your health, as well as spit acid from afar, so you should be extra careful. For each slain creature you will get 50 Glory Points. Note - don't stray off the main road, because to the East you can find a den of a huge Grave Spider - this foe can pose a serious threat to the inexperienced hero.

Connor

After reaching the Citadel, you can speak with the nearby Demon Hunters about the current state of Burke's, although its entirely optional and up to you. The most important thing to do here is to find a room, located on the ground floor, in which Connor runs his shop (point 1b on the map). During the conversation with Connor, he will speak about a magic formula needed to heal Burke - invest 300 gold to but it (don't worry though, as you will get back your investment quite soon). In the inventory of the Nameless Hero you will find a cure called Burke's Magic Formula.

Note - Connor is a dealer as well as a trainer, so don't be hasty with the conversation.

Burke is healed.

Speak with Burke and give him the magical formula (200 Glory Points). Leave the room and head to the Southern part of the Citadel. Locate the stairway leading to the upper floor of the building. After going through them, approach the healed Burke, located on the floor. The Demon Hunter will give you 400 gold for your efforts, meaning that you will earn 100 gold (the formula's cost was 300 gold).
