This page of the Uncharted 4: A Thief's End has a list of all optional conversations which serve as a separate category of secrets/collectibles. You will learn which chapters have optional conversations and how to unlock them.
Optional conversations are very easy to miss. The only way to activate a conversation like this is when an NPC has a dialog icon above their head. This icon can disappear if you move too far away or you miss it by having the camera facing the other way.

Conversations in chapter 3 - The Malaysia Job
During the third chapter you can initiate two optional conversations. The change to do so appears only during the final moments of the chapter, when you pull the trailer off the water and get back on the deck of the ship. Don't go to the meeting with Jameson yet, as you will end the chapter by doing so, Instead, look around the area for characters that you can talk to. You can distinguish them by the specific icons located above their heads. The ones you are looking are the workers - the one standing near the trailer (the first screenshot above), and the one standing on a small elevation (the second screenshot above).
Conversations in chapter 6 - Once a Thief...
During the sixth chapter of the game you can initiate three optional conversation with Nate's brother, Sam, who will be accompanying you during most of this section. You must be on your guard, as you can easily miss those moments. Like with the case of the third chapter, you will be informed about the opportunity to initiate a conversation when a specific icon appears above Sam's head (additionally, a characteristic sound will be heard). Opportunities to speak with Sam arise:
- After reaching the orchard.
- During the exploration of ledges, after the slide on the rope.
- During the exploration of the interior of the manor, when both characters are wearing suits - you must speak with Sam when he stands on a large external balcony (where you can find one of the secrets).
Conversations in chapter 8 - The Grave of Henry Avery
In chapter eight, you can have two optional conversations. The first conversation with Sam can take place when you investigate the three hanging crosses in the main chamber of the tomb. The second one can start once Sam lights a fire. Walk towards the locked door illuminated by sunlight, and only then talk to Sam.
Conversations in chapter 9 - Those Who Prove Worthy
There is only one optional conversation in chapter nine. Solve the turning mechanism puzzle, unlock the exit door and get to the viewpoint. Sam should stop and give you a chance to talk to him.
Conversations in chapter 10 - The Twelve Towers
There are two optional conversations that you can trigger in chapter ten, and both of them are in the same place. Get to a small tower on a hill; this will happen soon after you leave the biggest part of the map. Approach the tower and talk to Sam and Sully (this specific order).
Conversations in chapter 11 - Hidden in Plain Sight
In the eleventh chapter of the game, there only is one optional conversation. You will have it while crossing the bazaar. Approach the man with the lemur on his shoulder and pet the animal. Then, walk on. After a while, the conversation icon should appear above Sully's head and the conversation itself will concern Nate's fancy to get a pet for himself.
Conversations in chapter 12 - At Sea
In the twelfth chapter, there are two conversations that you can have with Sam. The first one may take place soon after he and Nate will have to split. After you reach, with Sam, the ledge that used to be inaccessible, you have to walk over, with him, to the nearby scenic point.
Another optional conversation takes place towards the end of this chapter, during the exploration of the pirate hideout. Before you enter the chamber with the puzzle (the second trial), you have to examine side rooms at the bottom level of the ruins. Start by examining the model in one of the rooms. Then, examine the figurine, two sketches and three paintings on the walls. Finally, swim down the narrow corridor to the neighboring chamber and examine the table there. Thanks to this, the conversation icon should appear above Sam's head (the screenshot).
Conversations in chapter 15 - The Thieves of Libertalia
The first one starts right after this chapter begins. Take several steps ahead and after a while, Sam will want to share his thoughts on the vault. After you start exploring the room, he will call you for the second time to have a conversation.
Conversations in chapter 16 - The Brothers Drake
After you climb up to the attic, take the ajar door into the room with toys. Pick up the letter on the desk. This will unlock the first optional conversation. The next one will take place after you enter the botanical room. Pick up the book about pirates and you can have two more optional conversations, thanks to this.
Conversations in chapter 17 - For better or worse
In this chapter, you can have six optional conversations. Right after the mission starts, when you go past the large flag with Elena, she will start the first one. Soon afterwards, you will start another conversation. Two more appear after you eliminate the two mercenaries. The last two take place towards the end of the mission, during the elevator ride.
Conversations in chapter 18 - New Devon
In this chapter, there are five optional conversations. First, show Elena around the starting location, next to the sign with the pirate motto, there is the first one. Another one takes place when Elena approaches one of the rocks Northwards of the sign. The third one takes place in the river that you drop into, after the bridge collapses. The last one takes place in the musical room in Avery's estate.
Conversations in the epilogue
Here, there is only one conversation. On the pier, you can have a "conversation" with your dog, which counts as the last optional conversation in the game.
