1. Completing main story of Mass Effect 2 should take you 25 hours, provided you play on normal difficulty level. In cases of higher difficulty levels, it may turn out that certain more difficult fights will have to be repeated. This may lengthen the time it requires to finish the story even by a few hours.
2. Completing all main and side quests takes about 35 hours. The game contains a lot of side quests, but some of them (e.g. N7 quests) are rather short. Companion quests are more demanding.
3. If you decide to explore the galaxy in 100%, the time elapsed increases to 50 hours. This means visiting all systems and scanning every planet.
4. Mass Effect 2 has a few large story DLCs. Achieving a 100% completion of each major DLC should take you around 2 hours. The lesser DLCs should take you around 30-60 minutes.
