Saints Row doesn't really describe Signature Abilities all too well. This page of the guide explains how to check the requirements of unlocking the vehicle or weapon's signature ability as well as how to activate them when equipped.

Vehicle abilities - unlocking and activating
Each vehicle in the game has its own signature ability you can unlock. It's displayed in the lower right corner of the screen. There are 10 different kinds of abilities available - some cars can share the same ability.
You unlock them by completing its challenge. These can force you to hit X cars with your towed cargo, hit X cars with Sideswipes or exhausting your boost X times. Completing some challenges requires mods purchasable while tuning the vehicle, such as the boost or the towing cable.
The game will let you know when you've beaten the challenge, though it doesn't automatically activate the ability.
First you must enter a garage and start tuning the car. At the bottom of the list there is an option to enable the vehicle's signature ability - do so. From now on you can use the vehicle's ability with the ability key (L1/LB on the controller). The abilities differ from each other. They can grant you unlimited boost, extending spikes for piercing tires and much more. You can read the ability's description in the garage, though we recommend trying it out by yourself.
Double Backflip - how to do?
One of the most problematic challenges in the game is doing double backflips. We recommend visiting the West Providencia cemetery to complete it.
There are a few large ramps there. Once you're in the air, tilt the stick down and don't let go. If you don't spend enough time in the air for a double backflip, equip a boost in the vehicle to remedy that.
Barrel Roll - how to do?
Another difficult challenge is doing an aerial barrel roll. The area marked on picture 1 in Marina East is a good place to complete it.
You'll find some large ramps here. Once you're in the air, hold down the drift button, then tilt the analog stick left or right. Your vehicle will start to flip. If you don't press the drift button, you'll start spinning instead, which won't complete the challenge for you.
Weapon abilities - unlocking and activating
Each weapon comes with its own unlockable signature ability. You can learn how it works and how to unlock it by highlighting it in the weapon wheel.
You unlock them by completing its challenge. These can force you to kill X enemies with your last mag, get X headshots or destroy X vehicles with the weapon.
The game will let you know when you've beaten the challenge, though it doesn't automatically activate the ability.
You must visit the armory at the Saints HQ or the Friendly Fire weapons store, highlight the ability and change it to active. Abilities have different effects, but they usually increase the gun's firepower or general effectiveness (sometimes only in specific circumstances, such as against vehicles).
